چکیده

Christian Baum ann Physiologisches Institut der Justus-Liebig-Universität Gießen, Aulweg 129, D-6300 Gießen, Bundesrepublik Deutschland Z. Naturforsch. 40c, 886—890 (1985); received October 23, 1985 Patch Clamp, Membrane ATPase, Tonoplast, Vacuole, Chenopodium rubrum L. The patch clamp technique has been applied to isolated vacuoles from green suspension cells of Chenopodium rubrum to record electrical parameters of the tonoplast. In a symmetrical K* solution of 46 mM, the membrane displays a near zero voltage, whereas 2 mM ATP will hyperpolarize it to 15 or 20 mV (vacuole positive). The conductance amounts to about one S-m~2. Fluctuations of the clamp current are explained by an unknown channel species having opening times of 5 — 10 ms. Together with previous work on a tonoplast vesicle preparation and unpub lished data on vacuoles from our laboratory, the present results indicate an electrogenic mem brane ATPase pumping protons from the cytoplasm to the vacuole.

ATP-dependent acidification and tonoplast hyperpolarization in isolated vacuoles from green suspension cells of Chenopodium rubrum L.

The tonoplast of isolated vacuoles from photoautotrophic suspension cells of Chenopodium rubrum L. was studied by means of the patch-clamp technique. In a symmetrical K(+) concentration of 46 mM, similar to in vivo conditions, the tonoplast displayed a membrane potential near zero and a linear current-voltage relationship with a mean slope of 1.0 S/m(2). ATP at 2 mM hyperpolarized the tonoplast...

Exoamylase activity in vacuoles isolated from pea and wheat leaf protoplasts.

Vacuoles isolated from pea (Pisum sativum), and wheat (Triticum aestivum) leaf protoplasts contained considerable activities of electrophoretically highly mobile exoamylases. Vacuoles from spinach (Spinacia oleracea) leaf and photoautotrophic Chenopodium rubrum suspension culture cell protoplasts were devoid of amylolytic activity. Endoamylase activity was in all cases associated primarily with...

Ultrastructure of the Shoot Apex of Chenopodium Album and Certain Other Seed Plants

The ultrastructure of cells of the vegetative shoot apices is described for Chenopodium album, Kalanchoë blossfeldiana and K. laxiflora, Bryophyllum daigremontianum, Nicotiana rustica, and N. tabacum (Maryland Mammoth), and Ginkgo biloba. A less intensive study was made of the last three listed. The structures and organelles usually associated with meristematic cells were observed: dictyosomes,...

Isolation of intact vacuoles and proteomic analysis of tonoplast from suspension-cultured cells of Arabidopsis thaliana.

A large number of proteins in the tonoplast, including pumps, carriers, ion channels and receptors support the various functions of the plant vacuole. To date, few proteins involved in these activities have been identified at the molecular level. In this study, proteomic analysis was used to identify new tonoplast proteins. A primary requirement of any organelle analysis by proteomics is that t...
